چکیده

1. The activity of erythrocyte 6aminolaevulinic acid (ALA) dehydratase and blood protoporphyrin concentrations have been measured in patients with various anaemias, a group of subjects with known lead exposure and a group of control subjects. Leucocyte ALA synthase was measured in subjects from the last two groups. 2. Erythrocyte ALA dehydratase activity was significantly depressed in the group of lead-exposed subjects and showed a highly significant negative exponential relationship with blood lead concentration. 3. Blood protoporphyrin concentrations were significantly elevated in the group of lead-exposed subjects and patients with iron-deficiency anaemia and showed a significant positive exponential relationship with blood lead concentration. 4. Comparison of the least-squares regression analysis of these relationships and incidence of false positive and false negative results indicates that erythrocyte ALA dehydratase activity is a more accurate measure of environmental and moderate industrial lead exposure than blood protoporphyrin concentrations. 5. The correlations of erythrocyte ALA dehydratase and leucocyte ALA synthase activity, and of blood protoporphyrin concentrations and leucocyte ALA synthase activity, suggest that blood

Influence of the degree of exposure to lead on relations between alcohol consumption and the biological indices of lead exposure: epidemiological study in a lead acid battery factory.

Alcohol has been shown to interact with lead to influence haem biosynthesis. The aim of this study was to define the dependence of this interaction on the degree of exposure to lead. Exposure to alcohol was estimated by measurement of alcohol concentrations in a sample of urine collected during the morning (AlcUM) (0.82 (SD 4.36) mmol/l) and in a sample collected during the afternoon (AlcUA) (1...

Effects of lead on haem biosynthesis during erythroid differentiation in vitro.

Murine erythroleukaemia cells (MELC) are erythroid precursor cells that undergo erythroid differentiation in the presence of the inducer hexamethylene bisacetamide (HMBA). The effects of lead on haem biosynthesis in MELC following HMBA-induced differentiation were studied. MELC were induced with HMBA in the presence of 20, 40 and 80 mum-lead acetate and cell density, haem content, incorporation...

EFFECT OF CHRONIC LEAD POISONING ON ERYTHROCYTE PROTOPORPHYRIN IN RATS

Research has shown that exposure to lead may have adverse effects at different blood lead concentrations. Lead inhibits at least two enzymes that are essential for the formation of heme, and because of the interaction of lead with these enzymes, no iron is inserted into protoporphyrin. Therefore the concentration of protoporphyrin increases in erythrocytes. The concentration of lead was mea...

Mobilisation of heavy metals into the urine by CaEDTA: relation to erythrocyte and plasma concentrations and exposure indicators.

To investigate the effects of calcium disodium ethylenediamine tetra-acetate (CaEDTA) on the urinary excretion, erythrocyte, and plasma concentrations and exposure indicators of seven heavy metals, CaEDTA was administered by intravenous infusion to 20 workers exposed to lead, zinc, and copper. The workers' blood lead concentrations ranged from 22 to 59 micrograms/dl (mean 38 micrograms/dl (1.8 ...

Lead toxicity and heme biosynthesis.

Lead intoxication results in a disturbance of heme biosynthesis, its degree depending on the severity and duration of exposure to lead. A mild secondary, sideroblastic anemia is common; basophilic stippling may occur, especially in severe lead poisoning. Increased excretion in the urine of delta-aminolevulinic acid and coproporphyrin III may occur; porphobilinogen excretion is not usually incre...
